The following experiment was aimed to clarify distributional change of the B lymphocyte and activated B lymphocyte after injection of the T lymphocyte independent B lymphocyte mitogen. Lipopolysaccharide (LPS), and T lymphocyte mitogen,
Phytohemmagglutinin (PHA). Male C57BL/6 mice were used in the experiment. The draining lymph modes of C57BL/6 mice were removed at 6, 12, 24 hours, 3, 7 days after the footpad injection of mitogens. Histochemical staining for the activated B
lymphocyte
and immunohistochemical staining such as avidin-biotin-peroxidase complex (ABC) were used. The distribution of activated B lymphocyte through measurement of membrane alkaline phosphatase (Apase) and the distribution of IgM positive B lymphocyte
were
observed.
@ES The results obtained from above experiment were as follows;
@EN 1. Normal control group : Surface IgM (sIgM) postive B lymphocyte were observed in peripheral cortex gererally, especially in lymph follicles, and activated B lymphocyte were distributed in medulla.
2) LPS injection group : sIgM positive B lymphocyte were observed obviously in the boundadry of deep cortex and medulla in 12 hors group and activated B lymphocyte were observed obviously in peripheral cortex in day group. In 3 and 7 days groups
after
injection, activated B lymphocyte increased in peripheral cortex and medulla.
3) PHA injection group : There was no difference between B lymphocyte and activated B lymphocyte distribution in PHA injection groups but size of B lymphocyte area decreased in 3 day group.
Above results suggest that the B lymphocyte activation and migration for mitogen stimulation are initiated in peripheral cortex, so activated B lymphocyte migrated through the boundary of the deep cortex and plasma lymphocyte appear in medulla
which
implies the termination.
